Overview:

Dimethorphan phosphate inhibits the cough center in the brain and is a non-addictive central antitussive. The effect is slightly better than dextromethorphan, about twice that of codeine, but the toxicity is lower, it will not become addictive after repeated use, and it is safer. It takes effect quickly after taking it, lasts for a long time, and does not cause constipation. There is no domestic manufacturer approved to market it, so it has great market potential. preparation:

Under nitrogen protection, add the starting materials 3-methoxy-17-methyl dextromorphan (5.71g, 20mmol) and bis(tricyclohexylphosphine)nickel dichloride (0.70g, 1mmol) into 100ml of toluene and stir Dissolve, raise the temperature to 65°C, dropwise add methylmagnesium bromide (tetrahydrofuran solution) (1mol/L, 20ml, 20mmol), complete the dropwise insulating reaction, and detect by TLC that the raw material reaction is completed. After the reaction is completed, the temperature is reduced to room temperature, and a small amount of saturated chloride is added dropwise. Quench the reaction with ammonium solution, add water to wash the layers, and separate the organic layer; the organic layer is washed with saturated brine, and then dried, and then the toluene is distilled off under reduced pressure to obtain dimethorphan; 25 ml of 95% ethanol is added directly without purification, stirred and dissolved at room temperature , then 85% phosphoric acid (2.3g, 20mmol) was added to react and a white solid precipitated. After filtration, washing and drying, 5.46g of dimethorphan phosphate was obtained with a yield of 77.4% and a purity of 96.2%.

Biological activity:

Dimemorfanphosphate is a potent agonist of sigma1receptor and can effectively relieve cough.
